The Protein-Pak SEC Column is shipped in 0.05% sodium azide in a water solution.

Resolve proteins that differ in molecular weight easily by a factor of two while distinguishing proteins that differ by as little as 15% in molecular weight. The degree of resolution depends more on the sample mass and volume than the interaction between the sample and the stationary phase. As secondary interactions are most often ionic, by increasing the ionic strength of the mobile phase, they can be reduced, for instance, by using salt concentrations in the 0.2-0.5M range (NaCl). In some cases, you can also use 10-20% methanol in order to eliminate hydrophobic and other hydrogen-bonding interactions.

To install the steel Protein-Pak SEC Column, start by removing the end plugs from your column with a 5/18” wrench and save them for storage when the column is removed from the system. The column outlet is indicated by an arrow on the label, which indicates the direction solvent should flow. Tighten the fittings 1/4-to-1/2 turn, but do not overtighten as this will damage the fitting seat. A properly prepared and assembled compression fitting in good conditions is all that is required.

What Sort Of Buffers Work With The Protein-Pak SEC Column?

The Protein-Pak SEC Column listed here is stable in normal salt buffer solutions such as sodium sulfate, ammonium acetate, ammonium formate, phosphate buffers, tris acetate, and acetate buffers. Ionic strength equivalent to 0.1 M minimizes the ionic interaction between the protein sample and the stationary silica phase. Generally, the salt concentrations in aqueous solutions should be maintained below 0.5 M.
