Spherisorb SAX Columns contain a silica-based quaternary ammonium bonded sorbent and are listed as USP L14 for use in anion-exchange separations. This column has a Parker-style end-fitting and a spherical particle shape. The inner diameter of each column is 4.6 mm and the length is 250 mm, with a particle size of 5 µm and a pore size of 80Å. Operating limits include a pH range of 2 pH to 8 pH and a maximum pressure of 6000 psi or 415 bar. It is recommended to use accessories of a matching chemistry such as a Spherisorb SAX Guard Cartridge, 80Å, 5 µm, 4.6 mm x 10 mm, 3/pk.

Each column is individually tested and shipped with a copy of test chromatograms produced during testing, including gel batch number, column serial number, USP plate count, USP tailing factor, capacity factor, and chromatographic conditions.

How Should I Use Guard Columns With Spherisorb SAX Columns?

Guard columns should always be of a matching chemistry and particle size between the injector and the main column. This is important as it protects the main column without compromising or changing the analytical resolution. Waters Spherisorb guard columns should be used. Guard columns should be replaced at regular intervals as determined by sample contamination. When system backpressure is observed as steadily increasing above a set pressure limit, this usually indicates the guard column needs to be replaced. Other indications of replacement needs are a sudden appearance of split peaks or other changes in chromatographic performance.
