How Can You Equilibrate Spherisorb Cyano (CN) Reverse Phase Column?
Spherisorb columns are shipped in test mobile phase; hence, it is important to ensure mobile phase compatibility before changing to a different mobile phase system. You can equilibrate the column by using a minimum of 10 column volumes of the mobile phase you intend to use. To ensure you avoid any precipitating out the mobile-phase buffers on your column, flush the column with five column volumes of a water or organic solvent mixture, using the same or lower solvent content as in the desired buffered mobile phase.
