Based on BEH technology the ACQUITY Amide columns offer a chemically stable, trifunctionally bonded amide phase that is stable from pH 2 to 12 to enable the separation of polar analytes that span a wide range of polarity, structural moiety and pKa.

Created to retain very polar acidic, basic, and neutral compounds, the ACQUITY UPLC BEH Amide Column overcomes common challenges in retaining and separating these compounds. Based on the ethylene bridged hybrid (BEH) particle technology, the columns use a chemically stable and trifunctionally bonded amide phase to enable enhanced stability and versatility in these separations. When used to retain polar analytes and metabolites that are too polar to retain through reversed-phase chromatography, the columns allow for the use of a wide range of mobile phase pH (2-11) to facilitate exceptional retention of polar analytes spanning a wide range of polarity, structural moiety, and pKa.

Along with enhanced retention of polar compounds, ACQUITY UPLC BEH Amide Columns offer increased mass spectrometry response, direct compatibility with sample preparation eluates (PPT, LLE, and SPE), and orthogonal selectivity when compared to reversed-phase materials.

Carbohydrates are one of the most abundant and diverse classes of compounds analyzed by HILIC, and the ACQUITY UPLC BEH Amide Column is an excellent tool for carbohydrate analysis. The 1.7 µm particle size allows for high resolution, high-speed analysis of carbohydrates in complex sample matrices while maintaining or improving chromatographic resolution. The addition of BEH particle technology provides superior column lifetime and improved assay robustness, which can be enhanced even further with the addition of lab equipment like the ACQUITY UPLC BEH Amide VanGuard Pre-column, 130Å, 1.7 µm, 2.1 mm X 5 mm, 3/pk.

The high pH mobile phase compatibility in ACQUITY UPLC BEH Amide columns also facilitates MS detection without the need for derivatization, post-column addition, or complexing with a metal cation. This dramatically simplifies sample pre-treatment before LC/MS analysis.

How Should I Prepare To Use My ACQUITY UPLC BEH Amide Column?

Columns will arrive in 100% acetonitrile. Before changing to a different mobile phase system, it is important to ensure mobile phase compatibility. Equilibration with a minimum of 10 column volumes of the mobile phase to be used is recommended. When a constant backpressure is achieved, the column can be considered thermally equilibrated. When your column arrives, flush it with 50 column volumes of a 60:40 acetonitrile:water combination with 10 mM final buffer concentration. Before the first injection is performed, equilibrate with 20 column volumes of initial mobile phase conditions.
