Hydrophilic-Interaction Chromatography (HILIC) is a technique used by chromatographers to retain and separate extremely polar compounds that could not otherwise be retained with more traditional routes like reversed-phase chromatography. This is also known as “aqueous mobile phase” or “reverse reversed-phase” because the elution order is that of a normal phase and solvents are similar to those of reversed-phase chromatography. The ACQUITY UPLC BEH HILIC Column uses Waters’ rugged 1.7 μm unbonded BEH particles in order to aid this retention and separation in very polar basic compounds.

These innovative columns are optimized and tested to provide the most reproducible separations possible under UPLC HILIC conditions. ACQUITY UPLC BEH HILIC Columns are designed to overcome the major weaknesses of lab equipment that uses HILIC stationary phases, focusing on chemical stability. While silica-based HILIC phases are often chemically unstable, the rugged BEH particle offers a wide usable pH range that overcomes this instability, resulting in long column lifetimes. Lifetimes can be prolonged using the ACQUITY UPLC BEH HILIC VanGuard Pre-column, 130Å, 1.7 µm, 2.1 mm X 5 mm, 3/pk.

As a part of the ACQUITY UPLC BEH family of columns, all packing materials are designed specifically for use with the ACQUITY UPLC system and are manufactured in the same cGMP, ISO 9001 certified plants with ultrapure reagents. Each batch of material undergoes chromatographic testing with acidic, basic, and neutral analytes, with results held to narrow specification ranges. Each column is also individually tested and includes a Performance Chromatogram and Certificate of Batch Analysis to help you ensure excellent and reproducible performance.

How Does HILIC Work?

HILIC (Hydrophilic-Interaction Chromatography) is a separation technique in which a polar stationary phase is used along with a mobile phase that contains a high concentration of non-polar (organic) solvent, and a low concentration of polar (aqueous) solvent. HILIC is used primarily when compound retention can’t be achieved through traditional reversed-phase liquid chromatography. HILIC has quickly gained popularity and a reputation as a powerful separation technique because the mobile phases used contain high concentrations of an organic solvent (such as MeCN) that are ideal for compound retention through ESI-MS. This allows for higher sensitivity and lower limits of detection in separations.
