PeptideWorks Tryptic Protein Digestion Kits uniquely deliver automatable, high efficiency, reproducible peptide maps in under 2.5 hours, 4x faster than average home-brew methods. With one kit, protein digestion can be performed manually or automated on Andrew+ Pipetting Robot and is amenable to other automated platforms. In addition, this kit contains a novel chemically optimized recombinant trypsin, RapiZyme Trypsin, Mass Spectrometry (MS) Grade for quick, clean and complete digestions in 30 minutes without contaminating autolysis peaks which can further complicate data processing. This starter kit includes everything needed for protein denaturation, reduction, alkylation, desalting and digestion to run a minimum of 4 samples up to 96 samples if needed. The kit provides: (2) bottles of Guanidine Hydrochloride (GuHCl) 5.7 g for denaturation, (8) bottles of Dithiothreitol (DTT) 16 mg for reducing, (8) bottles of Iodoacetamide (IAM) 53mg for alkylating, (12) x 8/pks of Sep-Pak SEC Desalting Cartridges with (1) 1 cc Cartridge Stand, (2) vials of 1M CaCl2 Salt Concentrate for desalting/digestion buffer and (4) vials RapiZyme Trypsin.
Customer will receive two packages. One package will ship cold-chain with all reagents and RapiZyme Trypsin. The second will ship ambient room temperature with the Sep-Pak SEC Desalting Cartridges and components.

The PeptideWorks Tryptic Protein Digestion Start-Up Kit is an all-in-one solution for scientists seeking rapid, high-efficiency protein digestion for peptide mapping workflows. With the ability to generate reproducible, high-quality peptide maps in under 2.5 hours, this kit is approximately four times faster than traditional in-house digestion methods. Whether used manually or integrated into automation platforms such as the Andrew+ Pipetting Robot, the kit is engineered to offer consistent performance across a variety of lab settings and throughput needs.
At the core of this kit is the innovative RapiZyme Trypsin—Mass Spectrometry (MS) Grade—a chemically optimized recombinant enzyme that completes clean and efficient digestions in just 30 minutes. Unlike traditional trypsins, RapiZyme minimizes contaminating autolysis peaks, enabling clearer MS data and streamlined downstream analysis. This enzyme is paired with all necessary reagents to support key steps such as protein denaturation, reduction, alkylation, desalting, and digestion.
The starter kit is equipped to process a minimum of 4 samples and can scale up to 96 samples, making it ideal for labs managing variable workloads. It includes: (2) bottles of Guanidine Hydrochloride (GuHCl) for denaturation, (8) bottles of Dithiothreitol (DTT) for reduction, (8) bottles of Iodoacetamide (IAM) for alkylation, (12) packs of 8 Sep-Pak SEC Desalting Cartridges with (1) Cartridge Stand, (2) vials of 1M CaCl2 Salt Concentrate, and (4) vials of RapiZyme Trypsin. This comprehensive layout eliminates the need for piecing together separate components and helps laboratories avoid delays.
The kit is delivered in two packages to maintain product integrity. One shipment arrives cold-chain with RapiZyme Trypsin and all temperature-sensitive reagents, while the other ships at ambient temperature with desalting cartridges and dry components. This ensures that both storage requirements and ease of setup are thoughtfully considered from the start.
Designed with flexibility in mind, the PeptideWorks Kit allows researchers to accelerate their peptide mapping workflows while ensuring reproducibility, compliance, and ease of integration into existing lab infrastructure. Whether you're exploring early-stage development or optimizing a validated process, this start-up kit simplifies protein digestion without sacrificing performance.

What makes the PeptideWorks Kit faster than traditional protein digestion methods?
The PeptideWorks Kit leverages RapiZyme Trypsin, a chemically optimized recombinant trypsin that delivers complete digestions in just 30 minutes. This streamlined enzyme activity, combined with a pre-validated reagent workflow, reduces the overall protein digestion time to under 2.5 hours—approximately four times faster than typical manual “home-brew” protocols. The reduction in steps and elimination of autolysis interference also helps accelerate data processing.
Is the PeptideWorks Kit compatible with automation platforms?
Yes, the kit is fully compatible with both manual workflows and automation platforms. It has been validated on the Andrew+ Pipetting Robot, but is also amenable to other automated liquid handling systems. This flexibility makes it ideal for both small-scale method development and high-throughput environments that require reproducible peptide mapping at scale.
What sample types and workflows are best suited for this kit?
The PeptideWorks Kit is optimized for peptide mapping applications involving biotherapeutic proteins, such as monoclonal antibodies or other recombinant proteins. It supports denaturation, reduction, alkylation, desalting, and enzymatic digestion—all essential steps in characterization and quality control workflows for biologics. The fast, automatable format is especially valuable in regulated environments where consistency and turnaround time are critical.
How should the PeptideWorks Kit components be stored upon arrival?
The kit arrives in two shipments. Reagents and RapiZyme Trypsin are shipped cold-chain and should be stored at -20°C immediately upon arrival to maintain enzyme activity. The Sep-Pak SEC Desalting Cartridges and related components ship separately at ambient temperature and can be stored in a dry location at room temperature. Proper storage ensures product stability and optimal performance across the full kit use range of 4 to 96 samples.
