Peptide mapping is a crucial process for proteomics and biotherapeutic characterization. It provides rich information on primary structure, modifications, and degradation of proteins. In proteomics, analysis is based on complex mixtures and partial sequence is used for protein identification with the aid of database searching tools. However, in the analysis of a well characterized biotherapeutic, there is a known sequence for individual proteins and a well defined list of modifications. For this type of study, full peptide mapping coverage is important in establishing comparability through formulation and process changes, determining stability, and investigating degradation pathways.
