We developed a novel sialylated glycopeptide enrichment method using two stage separations. In the first separation, a sample is extracted using an Metal Oxide Affinity Chromatography (MOAC) sorbent to selectively enrich sialylated glycopeptides. The enriched peptides are further separated and fractionated on a mixed mode HPLC column (strong cation exchange and reversed-phase/SCX-RP). The fractionated samples are deglycosylated and analyzed using NanoLC/MS/MS to identify the peptides with glycosylation sites.
