ABSTRACT: An ion-pair reversed-phase HPLC was used for analysis and purification of unmodified and fluorescently labeled oligonucleotides. Guidelines for column selection and optimization of mobile phase composition are discussed. Mobile phase consisting of triethylammonium-hexafluoroisopropanol ion-pairing buffer and methanol was directly compatible with mass spectrometry detection. The HPLC method was employed for quality control and purification of synthetic oligonucleotides and TaqMan oligonucleotide probes. NOTE: Search this database for Gilar for other related references.
