The patented RapiGest SF Surfactant is a reagent used to enhance enzymatic digestion of proteins, both in-gel and in-solution. RapiGest SF helps solubilize proteins, making them more susceptible to enzymatic cleavage without inhibiting enzyme activity.
Unlike other commonly used denaturants, such as SDS or urea, RapiGest SF does not modify peptides or suppress protease activity. It is compatible with enzymes such as Trypsin, Lys-C, Asp-N, Glu-C, PNGaseF, and other enzymes.
US Patent No: 7,229,539 and US 8,580,533
How RapiGest WorksRapiGest SF is a mild denaturant that solubilizes and unfolds the substrate protein, making it more amenable to cleavage. Mild protein denaturation opens the protein structure and exposes the proteolytic sites to enzymatic cleavage. More complex denaturation of globular proteins can be accomplished by heating in a RapiGest SF solution at elevated temperatures. |
Compatibility with MSRapiGest SF is an acid labile surfactant that undergoes hydrolysis in acidic conditions. This unique feature can be utilized to remove the surfactant from solutions when desired. Acidic conditions break down RapiGest SF to non-interfering by-products, therefore peptide ionization is not suppressed. |
The RapiGest SF AdvantageRapiGest SF shows significant advantage over other denaturants since it is not disruptive to endoprotease activity. It allows use of lesser amounts of expensive endoproteases. It is effective in a range of digestions such as Asp-N, Glu-C, Lys-C, PNGaseF and chymotrypsin digestions with the added advantage of significantly decreasing proteolytic digestion time, enabling same day digestions and analysis.
