Ion mobility separates molecules by <a href=\"https:\/\/ims.waters.com\/discover-ims-ms\/what-is-ccs\/\">size and shape<\/a> as well as m\/z. Cyclic IMS enables researchers to scale mobility resolution to match the complexity of their samples and performs unique IMS<sup>n<\/sup> experiments (e.g. selecting an ion by IMS, fragmenting the ion, selecting a particular fragment by IMS and repeating the process) that can reveal structural information that may never have been seen before.<\/p>\n<p>The combination of Electron Capture Dissociation (ECD) together with the more widely used Collision Induced Dissociation (CID), greatly enhances scientists\u2019 toolset for the analysis of complex biomolecules. ECD generally preserves the position of labile post-translational modifications and provides great sequence coverage for proteins and peptides.<\/p>\n<p>No less illuminating than structural characterization is knowledge about spatial localization. Following the time-honored truism that \u2018a picture paints a thousand words\u2019, <a href=\"https:\/\/vimeo.com\/420648939\/6ddb976cbf\">molecular maps can be generated using Desorption Electrospray Ionization (DESI)<\/a> \u2013 uniquely available on Waters\u2019 mass spectrometry systems. DESI XS is applicable to a broad range of different sample and surface types. Experiments can vary from tissue imaging in biomedical research and DMPK, through screening metabolites in bacterial colonies, to identifying fingerprints of different individuals taken directly from a surface and the examination of cross-sections of natural products like roots and tubers.<\/p>\n<blockquote><p>The new capabilities on the powerful Cyclic IMS platform contribute \u2013 through the elucidation of sequence, structure and spatial distribution &#8211; to a deeper understanding of molecular mechanisms of action.<\/p><\/blockquote>\n<figure id=\"attachment_8864\" aria-describedby=\"caption-attachment-8864\" style=\"width: 300px\" class=\"wp-caption alignleft\"><img decoding=\"async\" class=\"wp-image-8864 size-medium\" src=\"https:\/\/www.waters.com\/blog\/wp-content\/uploads\/ms-blog-image-300x170.jpg\" alt=\"\" width=\"300\" height=\"170\" \/><figcaption id=\"caption-attachment-8864\" class=\"wp-caption-text\">A high spatial resolution DESI-MS image of lipid species present in a sagittal mouse brain section<\/figcaption><\/figure>\n<p>Molecular visualization is even more comprehensive on Waters\u2019 discovery platform, the <a href=\"https:\/\/ims.waters.com\/the-synapt-xs\/\">SYNAPT XS<\/a>. Using DESI XS with complementary (optional) MALDI ionization maximizes the number and type of species whose distribution can be determined.<\/p>\n<p>Retaining high resolution imaging on a more accessible bench-top system, the Xevo G2-XS QTof also offers DESI XS technology; it not only accurately identifies \u00a0drug metabolites , for example, it can map their distribution across a tissue surface, giving scientists another dimension of information for developing the next generation of therapeutics.<\/p>\n<p>For routine analysis \u2013 where a molecule\u2019s characteristics have already been determined \u2013 the <a href=\"https:\/\/www.waters.com\/waters\/en_US\/Xevo-TQ-XS-Triple-Quadrupole-Mass-Spectrometry\/nav.htm?cid=134889751&amp;locale=en_US\">Xevo TQ-XS<\/a> and <a href=\"https:\/\/www.waters.com\/waters\/en_US\/Xevo-TQ-S-micro-Triple-Quadrupole-Mass-Spectrometry\/nav.htm?cid=134798856&amp;locale=en_US\">TQ-S micro<\/a> come into play. As a result of a collaboration with the University of Washington\u2019s Professor of Genome Sciences, <a href=\"https:\/\/www.maccosslab.org\/mike-maccoss\">Mike MacCoss<\/a> and his <a href=\"https:\/\/www.maccosslab.org\/skyline-team\">colleagues<\/a>, SkyLine software is now closely integrated with Waters MassLynx.<\/p>\n<p>The sensitivity of the Xevo TQ series combined with the industry standard <a href=\"https:\/\/skyline.ms\/project\/home\/software\/Skyline\/begin.view\">Skyline software<\/a> ensures that scientists in biomedical research or in pre-clinical bioanalysis in the pharmaceutical industry can routinely and accurately determine the quantity of their target molecules when developing peptide or protein-based therapeutics.
