Written by Waters scientist Jacquelyn Runco, this primer \u201c<a href=\"https:\/\/www.waters.com\/waters\/Beginner%27s-Guide-to-Preparative-SFC\/nav.htm?cid=134932612\"><strong>Beginner\u2019s Guide to Preparative Supercritical Fluid Chromatography<\/strong><\/a>\u201d introduces users to SFC, describes the technologies used in SFC analyses, provides practical tips and techniques, shows several practical examples from method development to scaling from analytical to prep.<\/p>\n<p>Chapters include:<\/p>\n<ul>\n<li>Introduction to SFx technology<\/li>\n<li>Introduction to prep SFC<\/li>\n<li>Enabling technology<\/li>\n<li>Diverse selectivity and applicability<\/li>\n<li>Method development<\/li>\n<li>Choosing a prep system<\/li>\n<li>Principals of scaling in prep SFC<\/li>\n<li>Example applications: Chiral and achiral purification<\/li>\n<li>Appendix A: CO<sub>2<\/sub> source<\/li>\n<li>Appendix B: CO<sub>2<\/sub> recycling<\/li>\n<li>Appendix C: Proper ventilation and lab monitoring<\/li>\n<\/ul>\n<blockquote><p>\u201cSFC is Chromatography!\u201d \u2013Jacquelyn Runco<\/p><\/blockquote>\n<p>As Jacquelyn enthusiastically writes, \u201cSFC IS CHROMATOGRAPHY! Supercritical fluid chromatography (SFC) is a chromatographic technique that uses sub-critical (liquid) and supercritical CO<sub>2<\/sub> as the primary solvent in the mobile-phase, usually accompanied by an organic solvent. Like all chromatography, SFC separates components based upon the partitioning of analytes between a stationary phase (column) and a mobile-phase (solvent).<\/p>\n<p>\u201cThere are many similarities between HPLC and SFC, for example: SFC can be run using both isocratic and gradient method conditions and is compatible with all standard detection techniques such as ultraviolet (UV), photodiode array (PDA), evaporative light scattering (ELS) and mass spectrometry (MS). \u2019<\/p>\n<p>\u201cThe general Prep SFC workflow is the same as for HPLC involving method development, scale-up, fraction collection and purity analysis of the collected fractions.
