We also hear the following question pretty often too:<\/p>\n<p><strong>VOC<\/strong>:\u00a0 <strong>\u201c<\/strong>I have a good analytical HPLC method, how do I scale it to prep?\u201d<\/p>\n<p><strong>Andy<\/strong>: In this short blog, I will provide some of the basic things to do when trying to scale an analytical method to prep.<\/p>\n<p>&nbsp;<\/p>\n<figure id=\"attachment_1734\" aria-describedby=\"caption-attachment-1734\" style=\"width: 350px\" class=\"wp-caption alignright\"><a href=\"https:\/\/www.waters.com\/blog\/wp-content\/uploads\/wg_blog_andyaubin_fig1.jpg\" rel=\"attachment wp-att-1734\"><img decoding=\"async\" class=\"wp-image-1734\" src=\"https:\/\/www.waters.com\/blog\/wp-content\/uploads\/wg_blog_andyaubin_fig1-300x181.jpg\" alt=\"WG_blog_AndyAubin_Fig1\" width=\"350\" height=\"211\" \/><\/a><figcaption id=\"caption-attachment-1734\" class=\"wp-caption-text\">Analytical Method<\/figcaption><\/figure>\n<h1><strong>Analytical Method<\/strong><\/h1>\n<p>First, develop the best analytical method possible. Better analytical methods mean better preparatory methods. Mobile phases need to be the same at both the analytical and preparative scales, identical A, identical B, and have same additive concentration. Samples need to be made at the same concentration using the same diluent. Ensure that a preparative version of your analytical column is available because your column chemistry needs to be the same.<\/p>\n<p>It also helps if both columns are of the same particle size and length. Loading studies can be helpful at this stage for users to confirm that adequate resolution will be maintained at the preparative scale.<\/p>\n<p>Finally, keep in mind that prep systems generally do not have column heating capability, so it might be better to develop your analytical method at room temperature.<\/p>\n<p>&nbsp;<\/p>\n<figure id=\"attachment_1736\" aria-describedby=\"caption-attachment-1736\" style=\"width: 350px\" class=\"wp-caption alignright\"><a href=\"https:\/\/www.waters.com\/blog\/wp-content\/uploads\/wg_blog_andyaubin_fig2.jpg\" rel=\"attachment wp-att-1736\"><img decoding=\"async\" class=\"wp-image-1736\" src=\"https:\/\/www.waters.com\/blog\/wp-content\/uploads\/wg_blog_andyaubin_fig2-300x183.jpg\" alt=\"WG_blog_AndyAubin_Fig2\" width=\"350\" height=\"214\" \/><\/a><figcaption id=\"caption-attachment-1736\" class=\"wp-caption-text\">Preparative Method<\/figcaption><\/figure>\n<h1><strong>Preparative Method<\/strong><\/h1>\n<p>Scaling your analytical method means adjusting your flow rate, gradient slope\/time (including compensation for system volume), and injection volume.
