This work demonstrates:

  • An efficient and generic 2D UPLC/MSE assay is developed for identification and quantification of host cell proteins (HCPs) in biotherapeutics that provides over four to five orders of magnitude in concentration.
  • This targeted, high-throughput quantitative assay for selected HCP proteins enables researchers to monitor their abundance changes.
  • Additionally, a high-throughput UPLC/MRM-MS assay for monitoring targeted HCPs using tandem quadrupole mass spectrometry can be readily implemented.
